You'll draft a team and have weekly matchups.
First, the game consists of a group of team "owners". A draft is scheduled for all owners to draft their "team" of individual players. All leagues are different in their settings. Some leagues have a salary cap to deal with where you have to stay in the parameter to field your roster. Some league's have IDP(Individual Defensive Players) formats. Others have a player slot to draft a collective team defense. Each roster can vary by league. Usually you have 2+ quarterbacks, 4+ running backs, 4+ wide receivers, 1 tight end, 1 kicker and a team defense. I am in one league where you have to draft an offensive line as well. Each team has a starting lineup for their weekly matchup. Usually, the owners play head-to head against each other on a weekly basis. Statistics are accumulated to figure out your individual lineups point totals. The team with the highest total in the matchup wins. There is a regular season then a playoff format. Usually the season runs through the first 14 weeks of the regular season in the NFL. The top few teams in the league make the playoffs and play for the championship. It is designed for fun but you actually have to do research on the players to field a good team. It gets competitive, from my personal experience, but it is a lot of fun. When this whole phenomenon began, players had to do everything on paper and it took a lot of work. With computers now, it is MUCH easier.
